Notes:The antibody used in this study by Wheatley et al., 1993 [PMID:7507029] was IM7. The method follows: "incubated with PBMT containing 10% foetal calf serum and a 1:50 dilution of mAb IM7 hybridoma supernatant overnight at 4�C followed by five 1 hour washes in PBMT. The primary antibody was detected by incubation in PBMT containing 1.6 mg/ml HRP-conjugated anti-rat Ig overnight at 4�C followed by washing as described above. For the enzyme reaction, the embryos were washed for 20 minutes in PBT (PBS plus 0.1% Triton X-100), incubated in PBT containing 0.08% NiCl2 and 250 mg/ml diaminobenzidine (Sigma) for 10 minutes and finally hydrogen peroxide was added to 0.03%." IM7 was from an immunization with the myeloid cell line M1.
