This is a traditionally produced wholemount in situ hybridisation stained using alkaline phosphatase + NBT/BCIP. It has been imaged using Optical Projection Tomography using visible light. In the OPT images, the signal is visible as the brightest regions.

Notes:The Wnt3a probe template used in this study was generated by RT-PCR from 9.5 embryo mRNA using the following primers: forward: 5'-AGAGCCAGGGTTGTTTCAGGA-3'; reverse: 5'-CTTTTGTCTAAATCCAGTGGTGGG-3', and this fragment was then cloned using a TOPO TA cloning kit (Invitrogen). Probe was transcribed from the entire insert.
