Notes:The Pcdh8 probe template used in this study by Makarenkova et al., 2005 [PMID:15627506] was "generated by genomic PCR. To detect both Pcdh8 isoforms (as used in this assay), a 1053-bp genomic fragment from the first coding exon was amplified (nucleotides 151-1203 from the initiation codon) using the following PCR primers: forward primer, 5' GAAGACCTGCACATGAAAGTGTTTGGAGAC 3'; reverse primer, 5' CAGCGAGGTGATACCGGCTTCATGTGTC 3'. This fragment was initially cloned into a TOPO vector (InVitrogen, Carlsbad, CA). It was then recloned as a HindIII-EcoRI fragment into pLITMUS 28 (New England Biolabs, Beverly, MA), and used as a template to generate a riboprobe by in vitro transcription using T7 polymerase".
