Notes:Runx1 expression was assayed in this study by Samokhvalov et al., 2006 [PMID:16496309] in embryos heterozygote for a targetted allele. A loxP flanked stop cassette was inserted downstream of the proximal promoter into the 5'UTR. To stop transcripts from the distal promoter, the cassette included a 1.7 kb splice acceptor sequence from the mouse engrailed-2 gene. The NLS was placed in-frame between the upstream distal promoter exons and the Betagal gene, and transcription stop elements were included downstream of lacZ to prevent read-through of the cassette. The neo was excised by transient expression of cre recombinase.
