This is a traditionally produced wholemount in situ hybridisation stained using alkaline phosphatase + BM Purple. It has been imaged using Optical Projection Tomography. The visible channel depicting the signal has been false-coloured yellow and the fluorescence channel depicting autofluorescence has been false-coloured blue.

Notes:Antisense probe sequence corresponds to coding and 3'UTR regions of the Dlx5 mRNA sequence. A Dlx5 fragment (1.6kb) was cloned into the pKS vector. The plasmid was linearised with Sal I and the antisense probe was transcribed with T3 RNA polymerase. Kindly provided by Rubenstein Lab; Ireland Lab of Developmental Neurobiology, San Francisco; constructed by Jen Kuei Liu. Probe available upon request.
