Notes:Efnb3 expression is detected in this study by Yokoyama et al., 2001 [PMID:11182083] in embryos heterozygous for a targeted mutation that was constructed as follows: Initially, a frt-flanked neo cassette and a loxP-flanked lacZ cassette were inserted at the endogenous locus. The neo cassette was inserted at a restriction site within intron 4 while the lacZ cassette was inserted in frame in exon 5, disrupting codons 264 through 340 which make up the majority of the cytoplasmic domain. The presence of the neo cassette prevented expression of lacZ. Then, the frt-flanked neo cassette was excised via FLP-mediated recombination, leaving the loxP flanked lacZ cassette in exon 5. The lacZ cassette disrupted codons 264 through 340 which make up the majority of the cytoplasmic domain. Biochemical analysis indicated that the expected EFNB2-beta-gal protein, in which the endogenous cytoplasmic domain is replaced with beta-gal, was produced. This mutation leaves forward signaling intact and ablates reverse signaling. The expression pattern of lacZ was observed to be to similar to that of the endogenous gene.
