Notes:
Mice homozygous for this targeted mutation are viable, fertile, and grossly normal but do exhibit a commissural defect, whereby tectal axons fail to project from the superior colliculus of the midbrain to the contralateral inferior colliculus and instead project to the ipsilateral cervical spinal cord. However, the authors show that at this stage, LacZ expression is more evident in the homozygote mutant, therefore the homozygote example is included in EMAGE. (A) Whole mount X-gal staining of (-/-) E10.5 embryo. LacZ expression is seen in the midbrain tectum (t), hindbrain (hb), rostral region of the spinal cord (s) and naso-lacrimal groove (n). No staining could be observed in wild-type littermates (not shown). (C) Vertical view (from above) of the E10.5(-/-) embryo depicted in (A).

Notes:Epha8 expression in this study was assayed in embryo heterozygous or homozygous for a targeted reporter allele where the exon encoding the extracellular domain was replaced with sequence encoding the transmembrane domain of the human NTRK1 receptor followed by lacZ and neo genes. The deletion was designed to generate a chimaeric protein in which the fibronectin type III-like domains were absent from the extracellular region, leaving only 116 carboxy terminal residues intact, and in which the entire cytoplasmic region was replaced by lacZ. LacZ activity was detected during embryonic stages and undetected during postnatal development.
