Fig2A shows a lateral view of wholemount stained 10.5dpc embryo. Figs3A and B show transverse sections of a 10.5dpc embryo after wholemount ISH. 3A shows the expression at the level of the maxillary and mandibular components of the first branchial arch, whereas 3B shows Gpc3 signals in the dermamyotome and sclerotome components of somites and no signals in the neural tube. Image annotations: in Fig2A: first (I) and second (II) branchial arches; limb buds (arrows), in Fig3A: mandibular (md) and maxillary (mx) components of the 1st branchial arch, in Fig3B: neural tube (nt), somintes (so).

Notes:The Gpc3 probe template used in this study by Pellegrini et al., 1998 [PMID:9853964] was generated using "primers derived from the human cDNA" where "an equivalent portion was recovered from a mouse embryonic library ("5'-stretch" from Clontech, Palo Alto, CA) by PCR amplification. The primers, which bear an EcoRI and XbaI site, respectively, at their 5' and 3' ends to facilitate cloning of the product, were CGGAATTCTTTCCAGTCATCTATACCC and GCTCTACACGAGTTCTTGTCCATTC. They amplify a 763-bp product (SGB.2) from the cDNA mixture."
