Notes:The Foxd4 (Fkh2) probe used in this study by Tamplin et al. was a gift from K. Kaestner, and indicated as that used by Kaestner et al., 1995 [PMID:8530406] . Further information from O. Tamplin summarising information from K. Kaestner defines the probe as "pg Fkh2 SalI/EcoRI 2.4, a 2.4 kb fragment of Fkh2 gene, contains entire coding region (only 1 exon) pBS II, KS-; T3 on 5' end (SalI); T7 on 3' end (EcoRI)". Editors note: Kaestner et al., 1995 describe three probes in their paper: probes A and B (both approx 0.2kb in length - see Fig 1 therein) which were fragments used for RNase protection probes, and probe C: a 2.6kb EcoRI-SalI genomic fragment used for screening a cDNA library). They do not indicate which fragment was used to generate ISH probe, however it is assumed that it was probe C as restriction enzyme analysis of the Foxd4 cDNA sequence X86368.1 (from Kaestner) identifies a 2.6kb fragment, and this fragment is identical to a genomic sequence from a SalI site just upstream from the single exon Foxd4 gene to a position 2.6kb downstream from it (where a linker derived EcoRI site is presumed to be situated in the pg Fkh2 SalI/EcoRI 2.4 (probeC) template clone).
