This is a traditionally produced wholemount in situ hybridisation stained using alkaline phosphatase + NBT/BCIP. It has been imaged using Optical Projection Tomography using visible light. In the OPT images, the signal is visible as the brightest regions.

Notes:The Wnt16 probe used in this study was generated by RT-PCR using the following primers: forward: 5'-ACGAGAGGTGGAACTGTATGG-3'; reverse: 5'-GCCTGCCAGAAAGAAAGAAATCG-3', and this fragment was then cloned using a TOPO TA cloning kit (Invitrogen). The probe was transcribed from the entire insert.
